    ProteoMiner technology is a novel sample enrichment tool for reducing the dynamic range of protein concentration in complex biological samples (serum, plasma and other sample types). Multiple kits are available to meet the needs of downstream applications:
    • Small-capacity kits ?optimized for limited sample material (minimum 10 mg of protein is recommended)
    • Large-capacity kits ?optimized for samples with 50 mg of protein

    PROTEIN SEPARATION


    2-D electrophoresis is used to separate complex protein samples based on pI and molecular weight. IEF is primarily used as the first dimension in 2-D analysis.

     

    2-D electrophoresis Liquid-Phase IEF

    The Protean IEF system, includes Protean IEF cell and ReadyStrip IPG, is optimized to perform first dimension IEF. This system offers a number of advantages:

    • streamlined handling,
    • enhanced resolution,
    • increased reproducibility

    The MicroRotofor cell is a preparative liquid-phase isoelectric focusing (IEF) device which fractionates microscale (~2.5 ml) protein samples into 10 fractions according to isoelectric point (pI) of protein samples in solution.

     

    Key Benefits

    • High-capacity running platform holds 1 large or 2 mini trays, allowing up to twenty-four 7cm strips per run or twelve 11, 17, 18 or 24-cm strips
    • Integrated 10,000 V power supply that helps to achieve desired volt-hours faster

    • Preset methods and real-time editing for maximum flexibility

    • Single- and multicolor fluorescent, chemiluminescent, chemifluorescent, and colorimetric samples

    • Calibrated transmissive and reflective scanning
    • 12-bit data and 36.3um resolution, allowing analysis of closest spots on a gel
    • Accurate quantitation of stained gels within a large dynamic range(0-3.0 OD)
    • Unique design to accomodate wet samples of variable thickness

    PD Quest software controls the imaging systems, collects and manages large amounts of image data, and rapidly and reliably analyzes the information.
    It facilitates the entire analysis workflow from image to identification.

    PDQuest combines:-
    1) Automated spot matching algorithms
    2) Normalization between gel images
    3) Statistical and qualitative query reports
    4) Merging of multiple images of the same gel

    TIPS AND TECHNIQUES


    Techniques such as 2-D electrophoresis (2DE) have been used for nearly 30 years to separate proteins. Like any other protein separation method, 2DE does have certain limitations in dealing with high molecular weight and membrane proteins. Bio-Rad has been in the electrophoresis business for years and helped customers answered many challenging questions related to 2DE analysis.
